Big Daddy.
You play as one of the prototype Big Daddies, one of the first ever made. The story behind it is that you were deactivated and then woken up ten years after the events of the first game. Upon being re-awoken, you find that you've regained your free will. Without spoiling much, you begin on a search for your original Little Sister from ten years ago(This is where story starts)
New edits to the big daddy role that you play:
You get to use plasmids and guns at the same time.
You can adopt or harvest little sisters. If you adopt, then you need to follow her to a corpse so she could extract Adam for you. When this occurs, you get ambushed from all directions by splicers.
Big Sisters are under the control of a woman who's trying to change Rapture. They protect little sisters and kill any splicer who's taken Adam from a corpse or Little Sister. You're their main enemy though because of the story line.

Actually, it's a slug.
The Little Sisters harvest a substance known as Adam from the people in the game. You can save/adopt them to collect Adam(Used to buy Plasmids[special abilities]) or you could kill them to access the actual slug and take the Adam from the little sister.
