QFT! I got a Virus once that did the same thing went onto google searching for a solution and smithfraud fix worked perfectly.

Use unlocker plus your AV. Find it and destroy it. Worse case, reformat, before that though try system restore.

at least find the name of the malware before you start installing anymore 3rd party software, which would cause more harm than good.

some are built to destroy specifided files and nothing more

It would be easier if you ran hijackthis log, and give us the log.

Do a hijackthis log and search for any supsicious BHOs.

BHOs are denoted by the prefix [O2].

Scan your computer with some Anti-Virus program (Maybe AVG Virus Free) and try to eliminate it via that program first. If not, if the virus's really strong, pehaps reformat. (Do reformat ONLY WHEN you really run out of ideas to get rid of it)
