once i set " Replace with code that does nothing " how can i patch or apply this modification to the exe or whatever i need to permanently modify with that code that does nothing ?

You want to permanently modify (corrupt) your game executable with the NOP?
Through CE, you could do File > Open File and select your EXE.
Find the code in there and NOP it. CE will overwrite the bytes in the file.

Make sure the game is unpacked before you waste time trying to find code that wouldn't exist yet
