I'm pretty sure you cant make a 16 bit be supported on a 64 bit system.
(unless you use emulation of some sort?)

I thought all AppData folders rebuild when a program runs or on reboot. I am not sure on this, maybe google it, but if its correct, try deleting the folder. To be honest i don't think it'll make a difference.

Don't delete the folder; it is a system folder so it could have unwanted consequences.

You may want to upload it to a site such as VirusTotal and see if it comes up with any results just in case.
http://www.virustotal.com/

If so you may want to either manually delete the file, or get a virus scanner that can remove the issue for you fully. (Manually removing it may not fix it entirely and the file could be replaced by another.)

Simple. There is some 16 bit application on your system that's trying to run at startup. Open msconfig (just type that into your start menu) and go to the startup tab. Find the program in the error message, and uncheck it. Problem solved, the program will no longer try to open, not be able to, and give you a message.

Hmm... I just checked inside the folder, but templatesloader.exe isn't there. Yesterday it was. How odd.

Try changing the folder permissions to see hidden files.

It's not hidden. I ran HijackThis and it showed none, but a "Loader.exe" for my sound driver. Possibly it's a Windows Update?

If it's a Windows update then they know a 16 bit exe won't run on 64 bit. Why would they include it? Go through your registry. Do virus scans, not just one use a few different products. Do as poster here suggested and run Hijack this and also run a BHO Demon. This application could be running in memory and only writing on winlogon start and stop. Then deleting itself.
