What is this paper club?
And what is this paper layout designer?
If it's for creating webpages then there are plenty of softwares to choose from.
But if it's for direct writing as in pen or pencil, I don't see how you can designed it that looks unique from those standard looseleaf formats.
What are you going to do?
Instead of a straight red line a inch from the left, you went a head and drew a curvy purple line instead?

Microsoft Visio is great for layouts, flowchart, and whatnot.
the alternative, and just as good, is OpenOffice Draw.
