Could be a malware that has disabled it.
Try this...
Go to registry editor (start run, type:regedit)
Navigate to:
HKEY_Current_User>Software>Microsoft>Windows>CurrentVersion>Policies>Systems
In the right pane, delete the value "DisableTaskMgr"
Restart
IF normal mode don't work, try it in safemode.

Here is a registry key, open notepad and paste the code in and save
as enableTaskMgr.reg
| Code: | Windows Registry Editor Version 5.00
[HKEY_CURRENT_USER\Software\Microsoft\Windows\CurrentVersion\Policies\System]
"DisableTaskMgr"=dword:00000000
|
Then double click on the registry file, restart your system and try again.

If you right click on the task bar can you access taskmanager from there?

Try Ctrl+Shift+Esc
Does task manager come up?

Hmm. not sure what the actual cause is since after looking at the web no one seems to have an direct answer. :\
Anyway tryout these options.
Go to device manager and remove the keyboard, restart, let system redetect keyboard and try again.
If not try the onsceen keyboard located in the accessibility menu

If it was disabled by a virus then try this...
