So can you actually use the pattern/signature scan to simply read values that are loaded into a register?

you can read bytes stored in memory (for example 000003A0), but in order to read a register then you need to debug it. (and i dont know how to debug an instruction and read registers contents)

You can write a cave to pull the value from the register and restore normal code flow etc. You can't read the register any other way besides debugging it as OldCheatEngineUser mentioned.

Another thing you can do though is trace back through the code to see if you can find where RDI is being set. If that is being set to a static pointer you can just use that +3A0 to get what you want.
