If i posted loads in random spam and lost about 50 post count. If i didnt post in RS for a week or two would my post count come back???

Nope. It's permanent.

You also have a chance of gaining posts also.

I got about 600 of my posts from RS.

It means that when you're posting in the Random Spam section, theres X chance of either getting some posts add'ed to your post count, or getting some posts removed from your post count..
